摘要:
为挖掘结直肠癌早期病变的生物标记物,通过对高通量组织芯片上进行优化的多重免疫组织化学染色,观察比对了TATA序列结合蛋白相关因子1 (TATA-box binding protein associated factor 1 like, TAF1L)和stomatin家族蛋白1(stomatin-like protein 1,STOML1)在结直肠癌组织和相匹配的癌旁正常组织之间的表达差异,检测了其表达水平与癌组织的不同临床分期、病理分级和Tumor-Lymph Node-Metastasis (TNM)分期的关系. 结果显示,相比癌旁正常组织,TAF1L蛋白质在癌组织中呈高表达,差异具有统计学显著性意义(P<0.000 1), 并可随原位病灶的恶化显现明显的表达改变.STOML1蛋白质的表达水平在癌组织中差异无统计学明显意义(P>0.05). 结果提示TAF1L蛋白质的异常高表达与结直肠癌的发生发展可能存在直接的关联,有可能成为结直肠癌早期诊断、精准治疗和预后判定的新分子标记物.
Abstract:
In order to obtain the therapeutic targets, protein expressions of TATA-box binding protein associated factor 1 like (TAF1L) and stomatin-like protein 1 (STOML1) on tissues are observed and analyzed via microarray of colorectal cancer and mulriple immunohistochemistry. Results show that protein expression of TAF1L is significantly increased in cancer tissues compared with paratumor tissues. Moreover, expression of TAF1L is closely linked to the process and malignancy of colorectal cancer. In contrast, protein expression of STOML1 is not statistically significant between cancer and paratumor groups (P>0.05). The conclusion suggests that TAF1L may be directly associated with the occurrence and development of colorectal cancer, and may also be as a potential biomarker for early diagnosis, precision medicine and prognosis to this disease.
