摘要:
阳离子纳米金是纳米金微粒与多聚L赖氨酸的结合物,在生理pH值条件下,标记在原代培养小鼠上皮成纤维细胞的阴离子位点.采用双光子荧光(two-photo fluorescence, TPF)显微和荧光寿命成像(fluorescence lifetime imaging microscopy,FLIM)技术,对阳离子纳米金标记的成纤维细胞进行二维和三维成像,可对小鼠皮肤重建过程中的成纤维细胞进行跟踪.
Abstract:
Fibroblasts were primary cultured from nu-nu/mice skin and grown in vitro on a glass surface or inside a dermal regeneration template matrix. 15 nm Cationic gold particles conjugated with poly-L-Lysine were attached on the anionic sites of fibroblasts on the membrane or inside the plasma because of their strong positive charge at physiological pH. Target cells labeled with cationic gold nanoparticles were excited by two-photon laser.Fluorescence and lifetime images were taken by confocal microscope and time correlated single-photon counting (TCSPC) module to verify the fluorescence of the matrix and the luminescence of the gold particles.Here the cationic gold nanoparticles labeled inside the fibroblasts acted as the tracer for tracking cells distribution in skin reconstruction.
