摘要:
通过设计合适的引物,经 RT-PCR 方法从里氏木霉的总 RNA 中扩增出 Swollenin 基因,并将该基因连接到 PMD18-T Simple 载体,得到该基因的克隆菌株,经酶切并将该基因连接到巴氏毕赤酵母表达载体 pPICZ α A ,得到重组质粒 pPICZ α A - swo1. 将该重组质粒线性化后,通过电转化方法转化毕赤酵母 GS115 ,得到重组菌株 P. pastoris-swo1. 该菌株的发酵液可使滤纸纤维强度降低,纤维素膨胀及崩解效果明显,表明重构表达的 Swollenin 可用于木质纤维素的预处理 .
Abstract:
The cDNA of swollenin gene was amplified from Trichoderma reesei by RT-PCR and doned in the PMD18- T Simple vector. The fragment of swollenin cDNA was then cut from PMD18-T Simple and ligated with the Pichia expression vector pPICZαA by restriction enzyme, resulting in the recombinant plasmid pPICZαA - swo1. The Linearized pPICZαA - swo1 was transformed into Pichia pastoris GS115 through electroporation, generating a recombinant strain which was named P. pastoris-swo1. P. pastoris-swo1 can reduce the tensility of filter paper fibre, and it swell and loosen cellulose obviously. Results indicate that the recombinant swollenin protein can be used in Pretreatment cellulose.
