摘要:
促性腺激素释放激素(GnRH)是在调控脊椎动物性腺发育和最后性成熟中起至关重要作用的一个保守神经十肽家族.从mGnRH被证实以来, 已经从原索动物和脊椎动物神经组织中发现16种不同形式的GnRH肽.在已研究的脊椎动物中, cGnRH-Ⅱ肽在所有脊椎动物脑组织中存在并完全保守,然而发挥主要功能作用的一般都是在下丘脑和端脑中优势表达的种属特异性GnRH肽.与脑组织中存在多种类型GnRH肽相对应的是, 在部分脊椎动物的垂体促性腺激素细胞膜上发现多种亚型的GnRH受体(GnRH-R).GnRH基因或GnRH-R的缺失或突变将导致生物个体繁殖生理学功能出现异常或病变.根据这一原理, 采用反义转基因技术, 通过在脑水平控制鱼类性腺发育, 已成功获得败育的转基因鲤鱼.采用该方法获得的转基因鱼在生态和遗传方面是安全的, 将为转基因鱼的产业化提供一种新的技术保障.
Abstract:
Gonadotropin-releasing hormone(GnRH),a conservative neuro-decapeptide family,plays a crucial role in regulating the gonad development and controlling the final sexual maturation in vertebrates. Since the first GnRH, namely mGnRH, was identified, 16 kinds of GnRH variants have been characterized from protochordate and vertebrate brain tissues to this day. cGnRH-Ⅱ peptide exist in all the brain tissue of detected organisms, and it is highly Conservative during the vertebrate evolution. However, the main function peptide is the GnRH, which is predominately expressed in hypothalamus and telencephalon. Corresponding to the multiplicity of GnRH peptide, different Subtypes of GnRH-R is also presented in gonadtropnn cell membrane of some vertebrates. So, it is proposed that the GnRH-R molecular have three kinds of subtypes which take form by homologous evolution.Thedeletion or mutant of GnRH gene or GnRH-R gene will lead to the abnormity or pathological changes of organism in dividual in propagate Physiology. According to the above theory, transgenic fish are produced using antiense transgenic technique, and Sterile transgenic fish are finally gained by inhibiting the expression of the sGnRH gene in brain level. Transgenic fish generated by antisense transgenic technology is safe in genetic and ecological aspects, which will supply a new technique scheme for the industrialization of transgenic fish.
