摘要:
ZNF219是新发现定位在人14号染色体q11带上的特异性锌指基因.本实验对其核定位信号的识别说明其中有两组核定位序列存在,因此通过将ZNF219的核定位序列(NLS)与N-端增强绿色荧光蛋白载体(pEGFP-N1)构建成质粒(pEGFP-ZNF219 NLS),然后瞬时转染到COS靶细胞,实验结果表明:绿色荧光蛋白(GFP)进入细胞核区.
Abstract:
ZNF219 is a novel zinc finger gene mapped to human chromosome 14q11..The cDNA of ZNF219 contains an open reading frame of 2169 nucleotides that encode 723 amino acids. The discrimination of nuclear localization signals showed that there are two sets of nuclear localization sequences (NLS) in it. A plasmid (pEGFP-ZNF219 NLS)was constructed by fusing the NLS of ZNF219 to a N-terminal enhanced fluorescent protein vector(pEGFP-N1),then transfected immediately to COS cell targeting the green fluorescent protein into the nucleus successfully.
