摘要:
以产胞内葡萄糖异构酶的树状黄杆菌(Flavobacterium araborescens) NRRL11022为出发菌株,用紫外线对其进行诱变,经筛选得到一株葡萄糖异构酶 的高产菌株U-616,其葡萄糖异构酶活力较出发菌株提高31%,经保存2年和多次 传代复测,其酶活力保持稳定.对诱变株U- 616进行产酶条件测试,结果发现其生长 和产酶需较高的溶氧水平,最适产酶温度为30℃,最适产酶pH为7.3 ~ 7.5,铁离 子对其生长和产酶无明显的影响,说明树状黄杆菌NRRL 11022的诱变株U-616 具有相当高的工业应用价值.
Abstract:
Flavobacterium Araborescens NRRL11022 was chosen as an original strain for screening high producer of glucose isomerase. After UV treatment, the mutant strain U-616 was selected. Its activity of glucose isomerase increased 31% over that of the parent strain. The enzyme activity was stable after eight times generation in two years. According to the test data of producing condition of glucose isomerase about the mutant U-616 strain it was found that high level of dissolved oxygen was necessary, the best enzyme producing temperature was 30℃ ,the best enzyme producing pH was 7.3 ?7.5,Fe2+ and Fe3+ did not have influence on cell growing and enzyme producing. Thus,the mutant strain U-616 had its potential value in industrial production.
