研究蛋白酶体激活因子REGγ对免疫细胞浸润能力的影响,分别腹腔注射李斯特菌于野生型和REGγ敲除小鼠,通过存活率检测、克隆形成、流式细胞术和苏木精-伊红染色实验研究,发现REGγ敲除小鼠表现为较低的存活率和李斯特菌清除能力.REGγ敲除显著抑制了中性粒细胞和单核细胞向感染部位的浸润.表明REGγ敲除后免疫细胞不能募集至感染部位,导致细菌未能被有效清除而在体内大量繁殖,最终引发败血症的发生,降低小鼠的存活率.

Peritoneal injections of Listeria Monocytogenes to wild-type and REGγ-knockout mice were performed for analyzing the roles of proteasome activator REGγ in immune cell infiltration. By survival assay, clone formation assay, flow cytometry and hematoxylin-eosin staining, we found that mice lacking REGγ shows significantly lower survival and Listeria Monocytogenes clearance. Furthermore, REGγ deficiency obviously inhibits the infiltration of neutrophil and monocyte to infection sites. Thus, the results suggest that REGγ deficiency suppresses immune cell recruitment to infection sites,resulting in impaired bacterial clearance. Rapid bacterial reproduction causes sepsis eventually, and leads to the reduced survival rate of REGγ-knockout mice.
