体外培养成骨MG- 63细胞, 通过倒置荧光显微镜和四甲基偶氮唑盐微量酶反应比色法,研究聚醚醚酮-磺化聚醚醚酮-羟基磷灰石(polyetheretherketone-sulfnated polyetheretherketone-hydroxyapatite, PEEK-SPEEK-HA)复合材料对细胞形态和增殖的影响.在PEEK-SPEEK-HA复合材料质量浓度为4 mg/mL时,复合材料对MG- 63细胞增殖作用最强,随浸提液质量浓度增大,材料对细胞增殖作用下降.材料质量浓度为4 mg/mL时,HA质量分数为20%的PEEK-SPEEK-HA复合材料对细胞的增殖作用较强. PEEK-HA和PEEK-SPEEK-HA复合材料的毒性评级为1级,无细胞毒性. SPEEK和HA能够改善复合材料对成骨MG- 63细胞的黏附作用,HA含量对细胞黏附作用的影响更显著. 扫描电镜结果表明MG- 63细胞在PEEK-SPEEK-HA材料上生长形态为三角形或长梭形,有伪足,伸展状态良好,细胞可以在材料缝隙处生长,表明成骨细胞能够在该材料表面黏附、伸展和生长. 将SPEEK引入PEEK-HA复合材料能抑制细胞凋亡.

Toxicity effects of PEEK-SPEEK-HA composite materials on the MG- 63 cells are studied in this paper. MTT results show that when the concentration of extracts increases, the cell proliferation decreases. In a concentration of 4 mg/mL,this material plays a significant role in cell proliferation. A concentration of 4 mg/mL has been selected to study the cell proliferation effect of different materials, and it is found that PEEK-SPEEK-HA(w(HA)=20%)composites have a stronger effect on cell proliferation than that of PEEK-HA(w(HA)=20%).The cytotoxicity of PEEK-SPEEK-HA and PEEK-HA composite is level 1, indicating no cytotoxicity. The cell adhesion rate increases with the increase of HA content in PEEK-HA composites, and scanning electron microscope(SEM)images show that MG- 63 cells can adhere, spread and grow on PEEK-SPEEK-HA surfaces. Apoptosis experiments show that PEEK-SPEEK-HA composite materials can inhibit apoptosis.
